Abstract
An #immunoenzymatic method for in situ detection or localization
of #abscisic acid (ABA) in cucumber leaves were developed and optimized.
Samples were sliced by frozen embedding and fixed in 2%
EDC(1-(3-Dimethylaminutesopropyl)-3-ethylcarbodiimide hydrochloride)
stationary liquid (3% paraformaldehyde, 0.5% #glutaraldehyde, 10mmol/LPBS
(phosphate buffer saline, pH 7.0) and in 20% sucrose for 1h. The
samples were then treated in 0.1% #trypsin for 5 minutes at 37 ℃. Sample
slices made by this method kept high integrity, the vein cells arranged
in order, peripheral cell wall was undamaged and the tissue keep the
most strong staining.
